How do you know which DNA strand is the template strand?

The template strand is one of the DNA strands whose base sequence helps in building mRNA through complementary base sequencing. Template strand or “Antisense strand” runs in 3′- 5′ direction, opposite to the coding strand.

Antisense is the non-coding DNA strand of a gene. A cell uses antisense DNA strand as a template for producing messenger RNA (mRNA) that directs the synthesis of a protein. Antisense can also refer to a method for silencing genes.

The 5′->3′ DNA strand is designated, for a given gene, as ‘sense’, ‘plus’ or ‘coding’ strand because its sequence is identical to the sequence of the premessenger (premRNA) [except for uracile (U) in RNA, instead of thymine (T) in DNA]. This coding strand is not transcribed .

The strand that contains the gene is called the sense strand, while the complementary strand is the antisense strand. The mRNA produced in transcription is a copy of the sense strand, but it is the antisense strand that is transcribed.

How is the template strand determine?

The RNA polymerase first attaches to the double helix of DNA and works with proteins called transcription factors to determine what information needs transcribing. The RNA polymerase and transcription factors bind to this DNA strand, called the template strand.

Is this strand of DNA considered coding or non coding?

When referring to DNA transcription, the coding strand (or informational strand) is the DNA strand whose base sequence is identical to the base sequence of the RNA transcript produced (although with thymine replaced by uracil). It is this strand which contains codons, while the non-coding strand contains anticodons.

Is the coding strand the sense strand?

The other strand is called the coding strand, because its sequence is the same as the RNA sequence that is produced, with the exception of U replacing T. It is also called sense strand, because the RNA sequence is the sequence that we use to determine what amino acids are produced through mRNA.

Is the positive strand the sense strand?

Depending on the context, sense may have slightly different meanings. For example, negative-sense strand of DNA is equivalent to the template strand, whereas the positive-sense strand is the non-template strand whose nucleotide sequence is equivalent to the sequence of the mRNA transcript.
